在改进姐妹染色单体区别染色(SCD)方法研究中,在对高压水银荧光灯的研究基础上,我们研制成灯泡内壁不涂以白色荧光粉的高压水银灯。用此灯对用5溴脱氧尿嘧啶核苷(BrdU)标记并用荧光染料双苯并咪唑(Hoechst33258,25μg/ml,PH6.8磷酸盐缓冲液配制),染色5—10分钟之染色体样品照射10~15分钟,再进行 Giemsa 染色。SCD 效果良好。这种方法之优点在于一灯兼起光源与热源之作用,代替了用普通紫外线灯照射外加水浴或其它加温方法的 SCD 步骤节省时间,提高 SCD 效果,自动计时使用方便;设备简单,价钱便宜。
Based on the research of high-pressure mercury fluorescent lamp, we developed a high-pressure mercury lamp with white fluorescent powder inside the inner wall of the bulb. Chromosome samples stained 5-10 minutes with a lamp labeled with 5 bromodeoxyuridine (BrdU) and fluorescently labeled bisbenzimidazole (Hoechst 33258, 25 μg / ml, pH 6.8 phosphate buffer) ~ 15 minutes, then Giemsa staining. SCD works well. The advantage of this method lies in the fact that a lamp combines the functions of a light source and a heat source to replace the SCD step of using an ordinary UV lamp to irradiate an external water bath or other warming methods to save time and improve the SCD effect, and the automatic timing is convenient; the equipment is simple and the price is cheap .
