本研究观察了杂色曲霉素(sterigmatocystin,ST)对小鼠Purkinje细胞TNF-α和TGF-β2 mRNA表达的影响。给予BALB/c小鼠一次灌胃ST(3000μg/kg),分别于灌胃后0.5、1、2、4、8和16h处死动物,用原位杂交方法观察了Purkinje细胞TNF-α和TGF-β2 mRNA的表达。TNF-α和TGF-β2mRNA在对照组和处理组各时间点的Purkinje细胞均有阳性表达。但各处理组与对照组比较,TNF-α和TGF-β2 mRNA表达显著增加(P<0.01)。上述结果说明单次ST灌胃后TNF-α和TGF-β2 mRNA在Purkinje细胞表达明显增加,TNF-α和TGF-β2在ST对脑组织的损伤过程中起着重要的作用。
This study investigated the effects of sterigmatocystin (ST) on the expression of TNF-α and TGF-β2 mRNA in mouse Purkinje cells. The BALB / c mice were given intragastric administration of ST (3000μg / kg), and animals were sacrificed at 0.5, 1, 2, 4, 8 and 16 hours after instillation. The in situ hybridization was used to observe the effects of TNF- β2 mRNA expression. The positive expression of TNF-α and TGF-β2 mRNA in Purkinje cells at different time points in control group and treatment group. However, compared with the control group, the expression of TNF-α and TGF-β2 mRNA increased significantly in all treatment groups (P <0.01). The above results indicate that the expression of TNF-α and TGF-β2 mRNA in Purkinje cells increased significantly after a single intragastric administration of ST, and TNF-α and TGF-β2 play an important role in ST injury of brain tissue.
