目的:优化金银花药材中木犀草苷的含量测定方法。方法:采用高效液相色谱法,以苯基硅烷键合硅胶为填充剂,色谱柱为Agilent ZORBAX SB-phenyl(4.6mm×250mm,5μm)柱,流动相为乙腈-0.5%醋酸溶液系统,梯度洗脱,流速1mL·min-1,检测波长350nm,柱温25℃,进样量10μL。结果:木犀草苷进样量在0.09~0.91μg范围内与峰面积呈良好的线性关系(r=0.9999);平均回收率(n=6)为103.1%,RSD为2.1%;检测限(S/N=3)为3.48ng,定量限(S/N=10)为8.70ng。结论:本法简便准确,可用于金银花药材中木犀草苷的含量测定。
Objective: To optimize the determination method of luteolin in honeysuckle. Methods: High performance liquid chromatography (HPLC) was used with phenylsilane bonded silica as the filler. The column was Agilent ZORBAX SB-phenyl (4.6 mm × 250 mm, 5 μm) with a mobile phase of acetonitrile-0.5% Elution, flow rate 1mL · min-1, detection wavelength of 350nm, column temperature 25 ℃, injection volume 10μL. Results: The calibration curve was linear in the range of 0.09-0.91 μg (r = 0.9999). The average recovery (n = 6) was 103.1% and the RSD was 2.1%. The detection limit / N = 3) was 3.48 ng and the limit of quantitation (S / N = 10) was 8.70 ng. Conclusion: This method is simple and accurate and can be used for the determination of luteolin in honeysuckle.
