目的:建立HPLC方法同时测定排石利胆片中大黄素、大黄酚含量的方法。方法:选择Agilent SB-C18(4.6mm×250mm,5μm)为色谱柱,流动相:乙腈-0.1%磷酸溶液(65:35),检测波长:254nm,进样量10μl,1.0m L/min流速,柱温为室温(25℃)条件下进行检测。结果 :大黄素、大黄酚分别在2.046ug·m L-1~51.15ug·m L-1、2.036ug·m L-1~50.90ug·m L-1范围内线性关系良好,平均回收率大黄素为100.02%,RSD1.6%,大黄酚为100.36%,RSD1.8%。结论 :采用高效液相色谱方法,可有效评估排石利胆片中大黄素及大黄酚的含量,本法简单、结果准确、灵敏度高。
OBJECTIVE: To establish a HPLC method for simultaneous determination of emodin and chrysophanol in Paishi Lidan tablets. Methods: Agilent SB-C18 (4.6mm × 250mm, 5μm) was used as the mobile phase. The mobile phase was acetonitrile-0.1% phosphoric acid solution (65:35). The detection wavelength was 254nm. The column temperature was measured at room temperature (25 ° C). Results: Emodin and chrysophanol had good linearity in the range of 2.046ug · m L-1 ~ 51.15ug · m L-1,2.036ug · m L-1 ~ 50.90ug · m L-1, respectively. The average recovery was rhubarb 100.02%, RSD 1.6%, chrysophanol 100.36%, RSD 1.8%. Conclusion: The HPLC method can effectively evaluate the content of emodin and chrysophanol in Paishi Decoction. The method is simple, accurate and sensitive.
