研究了抗人纤维蛋白单链抗体分子scFV-8E5与链亲和素融合蛋白的制备。链亲和素拼接在scFV-8E5 cDNA的3’端,制备的融合蛋白单体分子量约45kD。表达产物少数为周质间可溶型蛋白,多数为包涵体。无论是可溶型的蛋白产物或从包涵体中回收并复性的蛋白产物均有结合人纤维蛋白和生物素的功能。这个双特异性融合蛋白的成功制备证明了在scFV-8E5的3’端拼接其它效应分子的可行性。
Preparation of anti-human fibrin scFv-8E5 and streptavidin fusion protein was studied. The streptavidin was spliced at the 3 ’end of the scFV-8E5 cDNA. The prepared fusion protein had a molecular weight of about 45 kD. A small number of expression products for the periplasmic soluble protein, the majority of inclusion bodies. Both soluble protein products or protein products that are recovered and renatured from inclusion bodies have the function of binding to human fibrin and biotin. The successful preparation of this bispecific fusion protein demonstrated the feasibility of splicing other effector molecules to the 3 ’end of scFV-8E5.
