[目的]探讨知母总皂苷对Aβ25-35诱导PC12细胞凋亡的保护作用。[方法]用10~40μMAβ25-35分别刺激PC12细胞48h,MTT法测定细胞活力改变。分组:正常组,模型组,知母保护组。用0.5~40mg/L知母总皂苷和20μMAβ25-35共同作用PC12细胞48h,倒置相差显微镜观察各组细胞形态变化,MTT测其活力改变。[结果]MTT法显示不同浓度Aβ25-35可引起不同程度凋亡,成剂量依赖关系。不同浓度知母总皂苷保护PC12细胞,可抑制这种凋亡,5~20mg/L效果显著(P<0.01)。[结论]Aβ25-35能诱导PC12细胞凋亡,知母总皂苷对其诱导的细胞凋亡有明显保护作用。
[Objective] To investigate the protective effect of total timosaponins on Aβ25-35-induced PC12 cell apoptosis. [Methods] PC12 cells were stimulated with 10 ~ 40μMAβ25-35 for 48h respectively and the cell viability was measured by MTT assay. Grouping: normal group, model group, Anemarrhena asphodeloides group. The PC12 cells were treated with 0.5-40mg / L total timosaponins and 20μMAβ25-35 for 48h. The morphological changes of the cells were observed by inverted phase contrast microscope. The viability of the cells was measured by MTT assay. [Results] MTT assay showed that different concentrations of Aβ25-35 caused different degrees of apoptosis in a dose-dependent manner. Different concentrations of timosaponins protect PC12 cells from apoptosis, and the effect of 5 ~ 20mg / L is significant (P <0.01). [Conclusion] Aβ25-35 can induce the apoptosis of PC12 cells, and total timosaponin has a significant protective effect on its apoptosis.
