首次采用CO_2—SFE法提取洋金花中东莨菪碱,方法简便、快速、无环境污染,一个分析样品在二十分钟内即可萃取完成。并且建立了反相离子对HPLC法测定东莨菪碱的含量,方法专一、重现性好,加样回收率为98.78%,RSD=2.33%(n=3),日内变异和日间变异分别为0.95%和0.86%。系统考察了温度、压力、碱化剂、夹带剂、静态萃取时间、动态萃取量等条件对本法提取率的影响,确立了最佳萃取条件。
The CO_2-SFE method was used for the first time to extract the scopolamine from the golden flower. The method is simple, rapid, and free from environmental pollution. An analytical sample can be extracted within 20 minutes. The reversed-phase ion-pair HPLC method was established for the determination of scopolamine with specificity and reproducibility. The recovery rate was 98.78%, RSD was 2.33% (n=3), and the intraday variation and interday variation were 0.95. % and 0.86%. The effects of temperature, pressure, alkalinizer, entrainer, static extraction time, and dynamic extraction amount on the extraction rate of the method were investigated, and the optimal extraction conditions were established.
