背景与目的:研究c9,t11-共轭亚油酸(c9,t11-CLA)抑制人胃腺癌细胞(SGC-7901)中基质金属蛋白酶(MMP-2和MMP-9)的表达与亚油酸代谢途径中限速酶COX-2的关系。材料与方法:实验设200、100、50和25μmol/L的c9,t11-CLA4个剂量组,利用RT-PCR分别检测加入COX-2的选择性抑制剂NS-398前后SGC-7901细胞中MMP-2和MMP-9的表达。结果:未加入NS-398之前,25~200μmol/Lc9,t11-CLA均能抑制MMP-2和MMP-9mRNA的表达,与对照组相比,50、100、200μmol/L的c9,t11-CLA作用后,MMP-2和MMP-9mRNA的表达明显降低(P<0.01)。而加入NS-398使细胞中COX-2的活性被抑制后,c9,t11-CLA对MMP-2和MMP-9mRNA表达的抑制作用均消失,与对照组比较,25~200μmol/L的c9,t11-CLA作用后,MMP-2和MMP-9mRNA的表达均无明显变化(P>0.05)。结论:c9,t11-CLA能够抑制SGC-7901细胞中MMP-2和MMP-9mRNA的表达,这种抑制作用与COX-2有关。
BACKGROUND & AIM: To investigate the effects of c9 and t11-CLA on the expression of matrix metalloproteinase (MMP-2 and MMP-9) in human gastric adenocarcinoma cell line SGC-7901, Relationship between rate-limiting enzyme COX-2 and metabolic pathway. MATERIAL AND METHODS: Four doses of c9 and t11-CLA at 200, 100, 50 and 25μmol / L were prepared. The expression of MMPs in SGC-7901 cells before and after addition of COX-2 selective inhibitor NS-398 was detected by RT- -2 and MMP-9 expression. Results: Before adding NS-398, 25 ~ 200μmol / Lc9 and t11-CLA all inhibited the expression of MMP-2 and MMP-9 mRNA, compared with the control group, 50,100 and 200μmol / L c9 and t11-CLA After treatment, the expressions of MMP-2 and MMP-9 mRNA were significantly decreased (P <0.01). However, the inhibitory effect of c9 and t11-CLA on the expression of MMP-2 and MMP-9 mRNA disappeared when NS-398 was added into the cells. Compared with the control group, the c9, After the effect of t11-CLA, the expression of MMP-2 and MMP-9 mRNA had no significant changes (P> 0.05). CONCLUSION: c9 and t11-CLA can inhibit the expression of MMP-2 and MMP-9 mRNA in SGC-7901 cells. The inhibitory effect is related to COX-2.
