目的 了解健康中老年人外周血淋巴细胞热休克蛋白 70 (HSP70 )的基础表达情况及其是否可用于评估该年龄段人群在各种应激状态下对抗不良环境、疾病的能力。 方法 选择 10 2例健康中老年人 ,男性 5 2例 ,女性 5 0例 ,按年龄分为 40~ 49岁、5 0~ 5 9岁、60~ 69岁及 70~ 79岁4个组。分别抽取每人静脉血各 5ml,分离淋巴细胞 ,采用固定化蛋白质的免疫学测定 (Western印迹法 )检测其外周血淋巴细胞中HSP70。 结果 年龄与HSP70存在显著性相关关系 ,4个年龄组HSP70的含量 (积分光密度 )分别为 198 11± 77 72、168 3 3± 60 0 8、113 69± 3 4 12及 72 5 8±2 6 3 3 ,各组之间比较 ,差异具有显著性。建立“最优”回归方程为Y(HSP70 ) =3 64 81- 4 0 6X2 (年龄 ) ,即随年龄的增加HSP70表达逐渐降低。 结论 测定健康中老年人外周血HSP70的基础表达水平可作为该年龄段人群应激能力、健康状况的评价指标之一 ,用于评估人体在各种应激状态下对抗不良环境、疾病的能力
Objective To understand the basal expression of heat shock protein 70 (HSP70) in peripheral blood of healthy middle-aged and elderly people and whether it can be used to assess the ability of this age group to resist adverse environments and diseases under various stress conditions. Methods A total of 102 healthy and middle aged people were selected, including 52 males and 50 females. The patients were divided into 4 groups: 40-49, 50-59, 60-69 and 70-79 years old. Five ml of blood was collected from each patient and lymphocytes were isolated. HSP70 in peripheral blood lymphocytes was measured by immunological assay (Western blotting) using immobilized proteins. Results There was a significant correlation between age and HSP70. The content of HSP70 (Integral Optical Density) in the four age groups was 198 11 77 72, 16 63 3 60 80, 113 69 34 12 and 72 5 8 2 respectively. 6 3 3 , the difference between the groups was significant. The “optimal” regression equation was established as Y(HSP70) = 3 64 81- 4 0 6X2 (age), ie the expression of HSP70 gradually decreased with age. Conclusion The basic expression level of HSP70 in peripheral blood of healthy middle-aged and elderly people can be used as one of the indexes to evaluate the stress ability and health status of the people in this age group. It can be used to evaluate the ability of the human body to resist adverse environment and disease under various stress conditions.
