目的研究产前慢性缺氧对子代大鼠心脏结构的影响。方法 SD孕鼠25只,按电脑随机数字法均分为4个缺氧组(早期缺氧组、中期缺氧组、后期缺氧组、全期缺氧组)和空白对照组,每组各5只。出生子代大鼠分别于1 d龄、1月龄、3月龄、6月龄每组随机各取雌雄子鼠5只。监测缺氧对子鼠体质量、心脏质量的影响;检测子鼠心肌纤维直径;免疫组织化学方法测定心脏胶原Ⅰ、胶原Ⅲ的表达。结果产前慢性缺氧可导致仔鼠:(1)出生低体质量;(2)心脏质量指数增加;(3)6月龄时始出现心肌纤维直径增加(P<0.05),以妊娠中期缺氧组最明显。(4)6月龄时始出现心脏胶原Ⅰ、胶原Ⅲ的表达增加(P<0.05),以妊娠中期缺氧组最明显。结论产前慢性缺氧可致成年子代大鼠低出生体质量、心脏质量指数增加、心肌纤维增粗、心脏胶原表达增加。
Objective To study the effect of prenatal chronic hypoxia on cardiac structure in offspring rats. Methods Twenty-five SD pregnant rats were randomly divided into four groups: hypoxia group, hypoxia group, hypoxia group, hypoxia group and blank control group 5 only Five offspring of male and female rats were randomly selected from each of the first generation, the first month, the third month and the sixth month of age. The effects of hypoxia on the body weight and heart quality were monitored. The diameters of myocardial fibers were detected. The expressions of collagen Ⅰ and collagen Ⅲ were detected by immunohistochemistry. Results Prenatal chronic hypoxia could lead to pups: (1) low birth weight; (2) increased cardiac mass index; (3) myocardial fibrosis began to appear at 6 months of age (P <0.05) Oxygen group most obvious. (4) Cardiac collagen Ⅰ began to appear at 6 months of age, the expression of collagen Ⅲ increased (P <0.05), and was most obvious in the second trimester of pregnancy. Conclusion Prenatal chronic hypoxia can cause low birth weight, increased cardiac mass index, thickening of myocardial fibers and increased expression of cardiac collagen in adult offsprings.
