目的研究糖尿病大鼠局灶性脑缺血再灌注后血管他丁、基质金属蛋白酶-9在不同时间点的表达,探讨其病理生理机制。方法随机将Goto-Kakizaki(GK)、Wistar大鼠分为脑梗死组(CI组)、糖尿病脑梗死组(DM+CI组)及假手术组。采用线栓法建立大鼠大脑中动脉缺血再灌注模型,在不同的缺血再灌注时间点(3h、24h、3d、7d),采用Western blot法检测血管他丁蛋白表达、用明胶酶谱法检测基质金属蛋白酶-9(MMP-9)的活性。结果脑缺血再灌注后大鼠脑组织中血管他丁于再灌注3h开始升高,至24h达到高峰,随后逐渐降低,在24h时,DM+CI组较CI组增加显著(P<0.05);DM+CI组和CI组大鼠脑组织中MMP-9的活性较假手术组明显增加,DM+CI组明显高于CI组(P<0.01),且血管他丁的表达与MMP-9的活性存在直线相关关系。结论糖尿病缺血再灌注大鼠脑组织中的血管他丁的表达增加与MMP-9的活性相关,参与糖尿病合并脑梗死的病理生理过程。
Objective To investigate the expression of vascular statins and matrix metalloproteinase-9 at different time points after focal cerebral ischemia-reperfusion in diabetic rats and to explore the pathophysiological mechanism. Methods Goto-Kakizaki (GK) and Wistar rats were randomly divided into cerebral infarction group (CI group), diabetic cerebral infarction group (DM + CI group) and sham operation group. The rat middle cerebral artery occlusion (MCAO) model was established by thread occlusion. The expression of angiotensin-converting enzyme-linked immunosorbent assay was determined by Western blot at different time points of ischemia-reperfusion (3h, 24h, 3d, 7d) Method to detect the activity of matrix metalloproteinase-9 (MMP-9). Results The level of angiotensin in brain tissue of rats after cerebral ischemia and reperfusion increased from 3h after reperfusion to peak at 24h and then gradually decreased. At 24h, the DM + CI group increased significantly (P <0.05) The activity of MMP-9 in DM + CI group and CI group was significantly higher than that in sham-operation group, DM + CI group was significantly higher than CI group (P <0.01) There is a linear correlation between activity. Conclusion The increased expression of angiostatin in the brain tissue of diabetic rats with ischemia-reperfusion is related to the activity of MMP-9 and is involved in the pathophysiological process of diabetes complicated with cerebral infarction.
