【目的】通过对11批黄精煎剂中5-羟甲基糠醛含量的考察,为黄精的物质基础与质量控制研究奠定基础。【方法】采用煎煮法获得供试品溶液,高效液相色谱(HPLC)法测定其含量。色谱条件:HypersilODS2色谱柱(250mm×4.6mm,5μm),甲醇—体积分数0.05%磷酸溶液(体积比为10:90)为流动相,流速为1.0mL/min,检测波长为280nm。【结果】11批黄精煎剂中5-羟甲基糠醛含量的差别达30倍,在0.0200~2.0000μg(r=0.9997)范围内线性关系良好,平均回收率为100.3%(sR=1.9%,N=6)。【结论】该方法简便、结果准确,为黄精物质基础研究与质量控制提供了一定参考。
【Objective】 Through the investigation of the content of 5-hydroxymethylfurfural in 11 batches of Huang Jing decoction, it laid the foundation for the material basis and quality control of Huang Jing. 【Method】 The decoction method was used to obtain the test solution, and its content was determined by high performance liquid chromatography (HPLC). Chromatographic conditions: Hypersil ODS2 column (250 mm × 4.6 mm, 5 μm), methanol-0.05% phosphoric acid solution (volume ratio 10:90) was used as the mobile phase at a flow rate of 1.0 mL / min and a detection wavelength of 280 nm. 【Result】 The results showed that the difference of 5-hydroxymethylfurfural content in 11 batches of Huangjian decoction was 30-fold, with a good linear relationship in the range of 0.0200 ~ 2.0000 μg (r = 0.9997) with an average recovery of 100.3% (sR = 1.9% N = 6). 【Conclusion】 The method is simple and accurate. It provides a reference for the basic research and quality control of Polygonatum.
