目的研究新生SD大鼠星形胶质细胞培养上清液对神经干细胞(neuralstemcells,NSCS)体外定向分化的影响,探讨神经干细胞分化条件。方法收集新生和成年SD大鼠星形胶质细胞培养的上清液,以1∶3比例同DMEM/F12培基混合,分别对胎鼠来源的神经干细胞进行诱导分化,倒置相差显微境下观察细胞的生长情况,并采用免疫荧光检测方法对分化细胞鉴定和计数。结果新生鼠星形胶质细胞培养上清液与DMEM/F12的混合培基诱导神经干细胞分化为神经元的比例明显提高。结论新生鼠星形神经胶质细胞能促使神经干细胞向神经元方向分化。
Objective To study the effects of astrocyte culture supernatant of neonatal SD rats on the differentiation of neural stem cells (NSCs) in vitro and the differentiation conditions of neural stem cells. Methods Supernatants of astrocytes cultured in neonatal and adult SD rats were collected and mixed with DMEM / F12 in a ratio of 1: 3. The neural stem cells derived from fetal rat were induced to differentiate, The growth of the cells was observed, and the differentiated cells were identified and counted by immunofluorescence assay. Results The ratio of NSCs differentiated into neurons in neonatal rat astrocytes culture supernatant mixed with DMEM / F12 was significantly increased. Conclusion Astrocytes in neonatal rats can promote neural stem cells to differentiate into neurons.
