光镜下观察了正常雄性Wistar大鼠生后发育期胰岛胰多肽细胞(PP细胞)的形态和分布,并做计数与图像分析。生后1d和7d时PP细胞的突起不太明显;PP免疫反应物质较弱;并可在十二指肠上皮细胞间见到PP细胞。生后1d时PP细胞占有百分比很少,7d时开始增多,18d已达成年水平,28d时超过成年水平。这种数量变化主要表现在胰头部胰岛。PP细胞面数密度(NA)及体积密度(Vv)的变化与PP细胞百分数的变化基本一致。免疫电镜下观察到生后7d与28d及成年相比,PP细胞的分泌颗粒和金标记颗粒均较少。依PP细胞分泌颗粒核心电子密度和颗粒大小的不同,可见两种PP细胞:一种是分泌颗粒核心电子密度中等或空泡样,颗粒直径较大的为多;另一种是分泌颗粒核心电子密度较低且均匀,颗粒较小的为多。本实验提示大鼠胰岛PP细胞生后继续变化,并对上述变化的意义进行了探讨。
The morphology and distribution of islet pancreatic polypeptide (PP) cells in normal male Wistar rats were observed under light microscope and counted and imaged. The protuberances of PP cells were not obvious at 1d and 7d after birth. The PP immunoreactive substances were weaker. PP cells could be seen between duodenal epithelial cells. At 1 d after birth, the percentage of PP cells was small, increased at 7d, reached the adult level on the 18th day and exceeded the adult’s level on the 28th day. This change in quantity is mainly manifested in the pancreatic islets. PP cell number density (NA) and bulk density (Vv) changes and changes in the percentage of PP cells are basically the same. Immunoelectron microscopy showed that 7 days after birth and 28d and adult compared to PP cells secreting particles and gold-labeled particles were less. According to PP cell secretory particle core electron density and particle size of the different, we can see two kinds of PP cells: one is secretory particle core electron density medium or vacuole-like, larger particle diameter for more; the other is the secretion of particle core electron Lower density and uniform, smaller particles are more. This experiment suggests that the islet PP cells in rats continue to change after birth, and the significance of the above changes are discussed.
