目的:采用液体培养,研究发酵过程中光轮层炭壳菌菌丝体生长指标的动态变化情况。方法:分别采用考马斯亮蓝G-250法、紫外吸收法、氮蓝四唑(NBT)光化还原法、福林酚法,测定菌丝体蛋白质含量、过氧化氢酶(CAT)和超氧化物歧化酶(SOD)活性以及多酚含量。结果:在一定的发酵时间内,发酵液的p H无明显变化;菌丝体产量在第10天达到最大,为700 mg/100 m L;蛋白质含量最大可达到85.7 mg/g;CAT和SOD最高活性分别为44.11 U/g和68.74 U/g;多酚含量最大可达到2.65 mg/g。结论:建立光轮层炭壳菌菌丝体几种重要指标的动态变化,进一步了解其生长情况,为光轮层炭壳菌菌丝体的开发利用提供了重要依据。
OBJECTIVE: To study the dynamic changes of mycelial growth of C. carpio during the fermentation process by liquid culture. Methods: The contents of mycelium, catalase (CAT) and superoxide were determined by Coomassie brilliant blue G-250 method, UV absorption method, NBT photoreduction method and Folin phenol method respectively. Activity of dismutase (SOD) and polyphenol content. Results: During a certain fermentation time, the pH value of the fermentation broth did not change significantly. The mycelial yield reached the maximum at 700 mg / 100 m L on the 10th day, and the maximum protein content reached 85.7 mg / g. CAT and SOD The highest activities were 44.11 U / g and 68.74 U / g, respectively. The maximum polyphenol content reached 2.65 mg / g. Conclusion: The dynamic changes of several important indexes of mycelium of C. carpio were studied, and their growth was further studied. It provided an important basis for the development and utilization of mycelium of C. carpio.
