本研究利用甘薯徐薯18和徐781转录组测序结果,筛选到一条与淀粉合成相关的差异表达Unigene,编号为T1-30656。以徐781为材料,克隆该片段,并对其进行重测序和生物信息学分析。结果表明,该片段为果糖-1,6-二磷酸醛缩酶基因的c DNA序列,该基因包含3个外显子和2个内含子,亚细胞定位在细胞质当中,将其命名为Ib FBA5。该c DNA长为1 341 bp,含1 074 bp的开放阅读框,编码357个氨基酸。其序列与番茄、油菜、粳稻和拟南芥等相似性在77%~79%之间。
In this study, the sequencing results of sweetpotato Xuhu 18 and Xu 781 transcriptome were used to screen a differential expression Unigene related to starch synthesis, numbered as T1-30656. Using Xu 781 as material, the fragment was cloned and sequenced and analyzed by bioinformatics. The results showed that this fragment is the c DNA sequence of the fructose-1,6-bisphosphate aldolase gene, which contains three exons and two introns. The subcellular localization in the cytoplasm, named as Ib FBA5. The c DNA is 1 341 bp long and contains a 1,074 bp open reading frame encoding 357 amino acids. Its sequence similarity with tomato, rape, japonica and Arabidopsis, etc. in between 77% to 79%.
