目的:观察葡萄籽提取物(GSE)对小鼠移植性乳腺癌的抑瘤作用。方法:MTT法检测GSE在体外对小鼠乳腺癌细胞株的生长抑制作用;皮下注射法建立小鼠荷瘤动物模型,灌胃给药,分组为空白对照组、模型组及GSE50、100mg/kg组,灌胃给药5次/周,共4周;肿瘤病理切片做HE染色,并做免疫组化检测增殖细胞核抗原的表达情况,TUNEL试剂盒检测肿瘤组织凋亡情况。结果:(1)GSE在体外能抑制小鼠肿瘤细胞的增殖,作用有剂量依赖性(P<0.05)。(2)与模型组相比,50mg/kg和100mg/kgGSE组小鼠的肿瘤抑制率分别为34.78%和68.12%。(3)GSE对模型小鼠肝功能指标有一定改善作用。(4)免疫组化显示,GSE降低肿瘤组织中增殖核抗原PCNA的表达,同时TUNAL法检测肿瘤组织中凋亡细胞发现,GSE诱导肿瘤组织细胞发生凋亡。结论:葡萄籽提取物通过抑制肿瘤增殖和促进肿瘤组织的细胞凋亡而抑制小鼠乳腺癌细胞及荷瘤小鼠肿瘤的生长,且对荷瘤小鼠肝功能有一定的保护作用。
Objective: To observe the inhibitory effect of grape seed extract (GSE) on transplanted breast cancer in mice. METHODS: MTT assay was used to detect the growth inhibitory effect of GSE on mouse breast cancer cell lines in vitro. A mouse tumor-bearing animal model was established by subcutaneous injection and administered intragastrically. The groups were divided into blank control group, model group and GSE50, 100 mg/kg. The rats were intragastricly administrated 5 times per week for 4 weeks. The pathological sections of the tumors were stained with HE and immunohistochemistry was used to detect the expression of proliferating cell nuclear antigen. The TUNEL kit was used to detect the apoptosis of tumor tissues. Results: (1) GSE inhibited the proliferation of mouse tumor cells in vitro in a dose-dependent manner (P<0.05). (2) Compared with the model group, the tumor inhibition rates of the 50 mg/kg and 100 mg/kg GSE groups were 34.78% and 68.12%, respectively. (3) GSE has a certain role in improving the liver function of model mice. (4) Immunohistochemistry showed that GSE reduced the expression of proliferating cell nuclear antigen (PCNA) in tumor tissue, and TUNAL assay detected apoptotic cells in tumor tissue. GSE induced tumor cell apoptosis. CONCLUSION: Grape seed extract inhibits the growth of tumor cells in mice and tumor-bearing mice by inhibiting tumor proliferation and promoting tumor cell apoptosis, and has a protective effect on liver function in tumor-bearing mice.
