目的:研究敏咳清颗粒含药血清对经肝素刺激增殖的人胚肺成纤维细胞(lung fibrobast,LFB)表达纤维连接蛋白(fibronectin,FN)、干细胞因子(stem cell factor,SCF)的影响,探讨敏咳清颗粒治疗咳嗽变异性哮喘的作用机制。方法:将常规培养LFB细胞分为空白组、模型组、对照组、敏咳清高剂量组、敏咳清低剂量组、养阴清肺糖浆高剂量组、养阴清肺糖浆低剂量组、地塞米松组,加入肝素刺激后分别加入对应的含药血清,采用ELISA法检测各组细胞培养上清液中FN,SCF的水平。结果:①各组含药血清均可不同程度的抑制SCF的产生,敏咳清颗粒作用最强,并呈剂量依赖性;②敏咳清颗粒能明显抑制FN的表达,高、低剂量组之间相比无显著意义。结论:敏咳清颗粒能显著降低经肝素刺激增殖的LFB对FN,SCF的表达,提示抑制SCF,FN的表达,可能是其治疗咳嗽变异性哮喘(CVA)的作用机制之一。
Objective: To investigate the effect of Min Ke Qing Granules on the expression of fibronectin (FN) and stem cell factor (SCF) in human lung fibroblasts (LFB) stimulated by heparin, To investigate the mechanism of Min Ke Qing Granule in treating cough variant asthma. Methods: The conventional cultured LFB cells were divided into blank group, model group, control group, Min Ke Qing high dose group, Min Ke Qing low dose group, Yangyin Qingfei Syrup high dose group, Yangyin Qingfei Syrup low dose group, The rats in dexamethasone group were given heparin, and the corresponding drug-containing serum was added respectively. The levels of FN and SCF in the cell culture supernatants of each group were detected by ELISA. Results: (1) The serum of each group could inhibit the production of SCF to some extent, and Min Ke Qing Granules had the strongest effect in a dose-dependent manner. ② Min Ke Qing Granules could significantly inhibit the expression of FN, No significant difference between the time. CONCLUSION: Min Ke Qing Granule can significantly reduce the expression of FN and SCF induced by LFB stimulated by heparin, which may be one of the mechanisms of its treatment of CVA by inhibiting the expression of SCF and FN.
