用MTT法测定20种泰国植物乙醇提取物对斜纹夜蛾(Spodoptera litura)卵巢细胞系Sl-1的增殖抑制作用,用倒置相差显微镜观察细胞形态变化,用流式细胞术测定细胞线粒体膜电位变化。结果表明:5种植物(姜(Zingiber officinale Roscoe)、高良姜(Alpinia galangl(L.)Willd.)、印楝泰国变种(Azadirachta indica A.Juss var.siamensis Valeton)、姜黄(Curcuma longa Dennst)、泰国青柠(Citrus hystrix DC))乙醇提取物的细胞增殖抑制效果较好,抑制率明显大于阳性对照鱼藤酮,并且与提取物浓度成剂量依赖关系。不同提取物处理24h细胞形态发生明显改变,包括细胞贴壁性减弱、空泡化、细胞碎片产生;48h后细胞形态变化加强、空泡化严重、细胞碎片增多。5种植物提取物对细胞线粒体膜电位均有影响,其中印楝泰国变种、泰国青柠效果最好。初步认为姜、高良姜、印楝泰国变种、姜黄、泰国青柠对Sl-1有细胞毒性作用,值得进一步研究。
MTT assay was used to determine the inhibitory effect of 20 Thai plant ethanol extract on the proliferation of Spodoptera litura ovarian cell line Sl-1. The changes of cell morphology were observed by inverted phase contrast microscope. The changes of mitochondrial membrane potential . The results showed that five species (Zingiber officinale Roscoe, Alpinia galangl (L.) Willd.), Azadirachta indica A.Juss var.siamensis Valeton, Curcuma longa Dennst, Thai Citrus hystrix DC) ethanol extract showed better inhibitory effect on cell proliferation, and the inhibitory rate was significantly higher than that of the positive control rotenone, and it was dose-dependent with the concentration of extract. The morphological changes of cells treated with different extracts were significantly changed, including the decrease of adherent cells, vacuolization and cell debris generation. After 48 hours, the morphological changes of cells became stronger, the vacuoles became more serious and the number of cell debris increased. Five kinds of plant extracts on the mitochondrial membrane potential were affected, of which the Thai melon varieties, the best Thai lime. It is preliminarily believed that ginger, galangal, Neem Thailand variety, turmeric and thai lime have cytotoxic effect on Sl-1 and deserve further study.
