目的:通过琥珀酸脱氢酶(SDH) 电镜细胞化学的变化,探讨非创伤性下肢缺血预处理的心肌保护效应。方法:实验采用体重250 ±30g ,雄性SD 大鼠24 只,分4 组,每组6 只,即正常对照组( Ⅰ组) ;缺血再灌注组( Ⅱ组) ;经典缺血预处理组( Ⅲ组) ;非创伤性下肢缺血预处理组( Ⅳ组) 。按Nitro - BT 法显色制作SDH 光镜标本,按小川的亚铁氰化铜法制作电镜标本。光镜观察SDH 活性参照Niles 法分为4级。结果:Ⅰ组SDH 活性和心肌结构相对正常;Ⅱ组SDH 活性降低明显,线粒体损伤严重;Ⅳ组近于Ⅰ组;Ⅲ组界于Ⅰ组、Ⅳ组之间。结论:非创伤性下肢缺血预处理具有与经典缺血预处理相类似的抗心肌缺血再灌注损伤的效应。
OBJECTIVE: To investigate the myocardial protective effect of non-traumatic hypothalamic preconditioning by the change of cytochemistry in electron microscopy of succinate dehydrogenase (SDH). Methods: The body weight of 250 ± 30g and 24 male Sprague-Dawley rats were divided into 4 groups with 6 rats in each group, namely normal control group (group Ⅰ), ischemia reperfusion group (group Ⅱ), classic ischemic preconditioning group (Group Ⅲ), non-traumatic ischemic preconditioning group (group Ⅳ). According to Nitro - BT method, SDH light microscopy specimens were made and electron microscopy specimens were made according to the method of OCC. Light microscopy SDH activity according to Niles method is divided into 4 levels. Results: SDH activity and myocardial structure in group Ⅰ were relatively normal. SDH activity in group Ⅱ was significantly decreased and mitochondria were damaged severely. Group Ⅳ was more similar to group Ⅰ and group Ⅲ was in group Ⅰ and group Ⅳ. CONCLUSIONS: Noninvasive lower limb ischemic preconditioning has anti-myocardial ischemia-reperfusion injury effects similar to classical ischemic preconditioning.
