目的:研究评价三骨汤对骨转移癌OPG和RANKL表达的调节作用。方法:雌性SD大鼠左后肢胫骨的骨髓腔内接种乳腺癌MRMT-1细胞以制成乳腺癌骨转移模型。给予三骨汤治疗,以唑来膦酸为阳性对照。对胫骨标本用影像学方法进行评分,抗酒石酸酸性磷酸酶(TRAP)染色计数破骨细胞,免疫组化法检测OPG、RANKL表达并计算两者比值。结果:与模型组比较,中药组影像学评分改善趋势;TRAP(+)破骨细胞数量显著性减少(P<0.05),OPG表达极显著性增高(P<0.01),RANKL/OPG比值呈降低趋势。结论:三骨汤可有效抑制破骨细胞激活,减轻骨转移所致的溶骨性骨质破坏,其作用机制与促进OPG表达有关。
Objective: To study the effect of Sanji Decoction on the expression of OPG and RANKL in bone metastases. Methods: Breast cancer MRMT-1 cells were inoculated into the bone marrow of the left hind limb tibia of female Sprague-Dawley rats to make a model of breast cancer bone metastasis. Soup given soup treatment to zoledronic acid as a positive control. The tibia specimens were scored by imaging method, tartrate-resistant acid phosphatase (TRAP) staining was used to count the osteoclasts, and the expression of OPG and RANKL was detected by immunohistochemistry and the ratio of the two was calculated. Results: Compared with model group, the imaging score of TCM group improved; the number of TRAP (+) osteoclasts decreased significantly (P <0.05); the expression of OPG increased significantly (P <0.01); the ratio of RANKL / trend. Conclusion: Sanji Decoction can effectively inhibit the activation of osteoclasts and reduce osteolytic bone destruction caused by bone metastasis. Its mechanism is related to the promotion of OPG expression.
