目的:研究硫酸化苦豆子多糖(SAPS)溶液的构象特征以及在不同因素影响下的溶液行为,并考察其体外抗凝血活性。方法:采用刚果红法研究SAPS构象特征;通过圆二色谱(CD)研究SAPS在不同浓度、温度和添加钙离子等条件下构象的变化;并测定SAPS体外抗凝血活性。结果:刚果红试验中SAPS络合物的最大吸收波长会发生红移;浓度、温度变化以及添加钙离子使SAPS构象发生变化。不同取代度的SAPS表现出不同程度的抗凝血活性。结论:SAPS具有多股螺旋结构。浓度、温度和金属离子因素均对其构象均有不同程度的影响。随着硫酸根的取代度增加,SAPS的抗凝血作用增强。
OBJECTIVE: To study the conformational characteristics of SAPS solution and the behavior of solution under the influence of different factors, and to investigate its anticoagulant activity in vitro. Methods: The conformational characteristics of SAPS were studied by Congo red method. The conformational changes of SAPS under different concentrations, temperatures and addition of calcium ion were studied by circular dichroism (CD). The anticoagulant activity of SAPS in vitro was determined. Results: The maximum absorption wavelength of the SAPS complex in the Congo red test was red-shifted. The conformational changes of SAPS were observed when the concentration and temperature were changed and calcium ions were added. Different degrees of substitution of SAPS showed varying degrees of anticoagulant activity. Conclusion: SAPS has a multi-helix structure. Concentration, temperature and metal ions all have different degrees of conformation. The anticoagulant effect of SAPS is enhanced as the degree of substitution of sulfate is increased.
