基因编辑技术是研究基因功能的一个有效工具.CRISPR/Cas9系统是基于细菌Ⅱ型免疫系统改造而成的一种全新的基因编辑系统.因其相对于ZFNs、TALENs更加简单高效,适用于普通分子生物学实验室.各种植物特异的CRISPR/Cas9载体被构建并快速应用于多种植物中.本实验以棉花GhSBP基因为编辑对象,构建了CRISPR/Cas9系统敲除GhSBP基因的表达载体,并利用农杆菌介导法转化棉花,以期获得敲除GhSBP基因的棉花转基因株系.“,”Gene editing technology is a powerful tool to study the gene functions.The CRISPR/Cas9 system is a new gene editing system adapted from the prokaryotic type Ⅱ adaptive immunity system.Compared with ZFNs and TALENs,the CRISPR/Cas9 system was of more simplicity and higher efficiency,and it was applicable to ordinary molecular biology laboratory.Various plant-specific CRISPR/Cas9 vector systems have been established as the rapid adaption of this technology in many plant species.In this experiment,we used GhSBP gene of cotton as the target gene,constructed a CRISPR/Cas9 expression vector of knockout GhSBP gene,and then transformed cotton by A grobacterium-mediated transformation method to obtain transgenic cotton lines.
