目的:为了摸清肾综合征出血热(HFRS)病毒对人血管内皮细胞(HECs)骨架的作用.方法:采用细胞培养、细胞化学和免疫细胞化学染色及光镜和电镜等技术,对HFRS病毒体外感染的HECs进行观察和研究.结果:发现病毒感染的内皮细胞骨架蛋白含量和分布有改变,致密周围带微丝和外周带中等纤维破坏和消失,导致内皮细胞回缩,细胞间裂隙形成.细胞形态改变的程度与所接种病毒的滴度呈正相关.结论:HFRS病毒的感染可改变HECs的细胞骨架的结构、分布和功能,进而导致血管壁通透性增加.
Objective: To find out the effect of hemorrhagic fever with renal syndrome (HFRS) virus on the skeleton of human vascular endothelial cells (HECs). Methods: HECs infected with HFRS virus in vitro were observed and studied by cell culture, cytochemical and immunocytochemical staining, light microscopy and electron microscopy. Results: It was found that the contents and distribution of skeletal proteins in endothelial cells were changed. The microfilaments and the peripheral fibers in the dense zone were destroyed and disappeared, resulting in the contraction of endothelial cells and the formation of intercellular fissures. The degree of cell morphology change was positively correlated with the titer of the inoculated virus. Conclusion: Infection with HFRS can change the structure, distribution and function of cytoskeleton in HECs, leading to increased permeability of the vascular wall.
