用60Co-γ射线辐射小麦品种河科2号的干种子,对M4代234个株系的蛋白质含量、湿面筋含量、沉降值、硬度的变异进行了分析。以超过群体均值±2×标准差为选择标准,筛选出6个蛋白质和湿面筋含量正向变异株系和3个负向变异株系。在M5代对它们的粉质参数进行了测定,并利用SDS-PAGE对其高分子量麦谷蛋白亚基(HMW-GS)进行了鉴定。结果表明,株系242的粉质图谱与粉质参数显著改变,株系240、108的HMW-GS组成也存在明显差异。亲本的HMW-GS组成为2+7+8+9+12,株系240、108的HMW-GS组成为2+7+8+12,说明在麦谷蛋白位点上发生了变异。
The variation of protein content, wet gluten content, sedimentation value and hardness of 234 lines of M4 generation were analyzed by using 60 Co-γ ray to irradiate the dry seeds of Heke No.2. With the standard of ± 2 × standard deviation of the population mean value as the selection criterion, six protein and wet gluten content positive mutant strains and three negative mutant strains were screened out. Their silty-forming parameters were measured in M5 and their high molecular weight glutenin subunits (HMW-GS) were identified by SDS-PAGE. The results showed that the silty diagram and silt parameter of strain 242 changed significantly. There were also significant differences in HMW-GS composition between strains 240 and 108. The HMW-GS composition of the parents was 2 + 7 + 8 + 9 + 12, and the HMW-GS composition of lines 240 and 108 was 2 + 7 + 8 + 12, indicating a variation in glutenin sites.
