目的:评价蛇床子总香豆素(total coumarins from dried fruits of Cnidium monnieri,TCCM)抗骨质疏松的剂量反应,并与尼尔雌醇(nilestriol,Nil)比较.方法:用3月龄去卵巢SD雌性大鼠,TCCM67 mg·kg~(-1)和200 mg·kg~(-1) ig,均每周6次;Nil1mg·kg~(-1)ig,每周1次,持续12 wk.取胚骨,不脱钙骨片测量.结果:去卵巢大鼠骨小梁面积下降59%,出现骨吸收大于骨形成的高转换型骨质疏松;低剂量的TCCM仅部分抑制骨高转换,不能阻止骨质疏松的发生(与同年龄组大鼠相比骨小 梁面积减少43%);高剂量的TCCM和Nil(比去卵巢组骨小梁面积分别增加100%和274%)一样,完全抑制骨高转换,能有效防止去卵巢后骨质疏松,但Nil抑制骨高转换的作用较强.结论:高剂量TCCM通过抑制骨转换对去卵巢大鼠有预防骨质疏松的作用,而Nil作用较TCCM强.
Objective: To evaluate the dose response of total coumarins from dried fruits of Cnidium monnieri (TCCM) against osteoporosis and to compare with nilestriol (Niltriol) .Methods: SD female rats were treated with TCCM 67 mg · kg -1 and 200 mg · kg -1 ig every 6 times a week and Nil 1 mg · kg -1 once a week for 12 weeks .Bone trabecular bone was taken and the non-decalcified bone fragments were measured.Results: The trabecular area of ovariectomized rats decreased by 59%, and high conversion osteoporosis with greater bone resorption than bone formation was found. Low-dose TCCM only partially inhibited bone turnover , Failed to prevent osteoporosis (43% reduction in trabecular area compared with rats in the same age group); high doses of TCCM and Nil (trabecular area increased by 100% and 274%, respectively, in the ovariectomized group) , Completely inhibited the conversion of bone height, can effectively prevent osteoporosis after ovariectomy, but Nil strong inhibition of bone turnover role.Conclusion: High-dose TCCM can prevent osteoporosis in ovariectomized rats by inhibiting bone turnover, Nil is stronger than TCCM.
