本文报道了从牛气管中分离及提纯硫酸软骨素A(ChS-A)及硫酸软骨素C(ChS-C)的方法。为了除去与硫酸软骨素结合的蛋白质并尽量减少硫酸软骨素的降解,我们采用了两种蛋白质水解酶,即胰蛋白酶及木瓜蛋白酶,使大部分的蛋白质分解。然后再采用有机溶剂沉淀法,活化浮石土吸附法以及离子交换树脂等,以去除少量的残余蛋白。加入氯化十六烷吡啶使ChS-A及ChS-C沉淀,并利用此沉淀在不同浓度的氯化钠溶液中溶解度不同的这一特性以去除可能与ChS-A及ChS-C同时存在的其它酸性粘多糖。最后用乙醇分段沉淀法将ChS-A及ChS-C分开。用本法所提制出的ChS-A及ChS-C经化学及物理化学分析证明纯度很高。
In this paper, a method to separate and purify chondroitin sulfate A (ChS-A) and chondroitin sulfate C (ChS-C) from bovine trachea is reported. In order to remove the protein bound to chondroitin sulfate and minimize the degradation of chondroitin sulfate, we used two proteolytic enzymes, trypsin and papain, to decompose most of the protein. Then, organic solvent precipitation method was used to activate pumice-soil adsorption method and ion exchange resin to remove a small amount of residual protein. Hexadecylpyridine chloride was added to precipitate the ChS-A and ChS-C, and this precipitate was used in different concentrations of sodium chloride solution with different solubility characteristics to remove possible simultaneous presence of ChS-A and ChS-C. Other acid mucopolysaccharides. Finally, ChS-A and ChS-C were separated by ethanol fractionation. The purity of ChS-A and ChS-C produced by this method was confirmed by chemical and physical chemical analysis.
