目的:观察新型壬苯醇醚(NP-9)膜体内、外杀精作用和家兔抗生育效果。方法:①体内、外杀精试验:进行体外和家兔阴道内不同浓度杀精剂的杀精试验,观察其的最低有效浓度。②家兔抗生育实验:按照在动情期投入不同的药物,将家兔随机分成3组,空白组不给药物与雄兔自然交配,阳性对照组在给予市售NP-9膜10 min后与雄兔交配,实验组在给予新型NP-9膜后立即与雄兔交配,计算妊娠率和半数有效量(ED50)。结果:①新型NP-9膜组溶液中NP-9体外最低有效杀精浓度为0.35 mg/ml,NP-9膜组溶液中NP-9和NP-9原料组体外最低有效杀精浓度均为0.25 mg/ml NP-9,新型NP-9膜体内杀精最低有效浓度为1 mg/kg NP-9。②家兔妊娠率空白对照组为100%,10 mg/kg NP-9阳性对照组为0%,1 mg/kg NP-9实验组为0%。实验组ED50为1.1771 mg/kg,低于阳性对照组(3.7931 mg/kg),P<0.01。结论:新型NP-9膜体外最低有效杀精浓度比壬苯醇醚膜组高,但体内杀精效果高于NP-9膜;而ED50低于NP-9膜,显示新型NP-9膜有良好的应用前景。
Objective: To observe the in vitro and in vivo spermicidal effects of a novel nonoxynol (NP-9) membrane and the anti-fertility effect of rabbits. Methods: ① In vitro and in vivo spermicidal tests: The spermicidal test of different concentrations of spermicide in vitro and in rabbits was carried out to observe its minimum effective concentration. Rabbit antifertility experiment: In accordance with the investment in estrus during different drugs, the rabbits were randomly divided into three groups, the blank group did not give the natural mating with male rabbits, the positive control group after the commercially available NP-9 membrane 10 min and The male rabbits were mated. The experimental group was mated with the male rabbits immediately after the administration of the new NP-9 film, and the pregnancy rate and the median effective dose (ED50) were calculated. Results: ① The minimum effective spermicidal concentration of NP-9 in NP-9 membrane was 0.35 mg / ml in vitro. The lowest effective spermicidal concentrations of NP-9 and NP-9 in NP-9 membrane were 0.25 mg / ml NP-9, the lowest effective concentration of NP-9 membrane spermicide was 1 mg / kg NP-9. ② The pregnancy rate in rabbits was 100% in blank control group, 0% in 10 mg / kg NP-9 positive control group and 0% in 1 mg / kg NP-9 experimental group.
