对香蕉束顶病毒(BBTV)中国分离株DNA组份I(DNA1)、外壳蛋白(CP)和运转蛋白(MP)基因进行了克隆和序列分析。BBTVDNA1含有1103个核苷酸,与南太平洋和亚洲分离株分别有87%88%和96%98%的核苷酸序列同源性。由DNA1编码的复制酶含有186个氨基酸残基,与南太平洋和亚洲分离株分别有944%958%和976%98.0%的氨基酸序列同源性。外壳蛋白基因由510个核苷酸组成,与澳大利亚分离株有925%的核苷酸序列同源性和988%的氨基酸序列同源性。运转蛋白基因由348个核苷酸组成,与澳大利亚分离株有906%的核苷酸序列同源性和879%的氨基酸序列同源性。通过比较中国分离株和其它国家分离株的DNA1、CP和MP基因的核苷酸序列,进一步证明BBTV分离株明显可分为南太平洋和亚洲两组。但是两组分离株CP基因的氨基酸序列是很相似的。
The DNA components I (DNA1), coat protein (CP) and prolactin (MP) genes of Chinese banana bunchy top virus (BBTV) were cloned and sequenced. BBTVDNA-1 contains 1103 nucleotides, which share 87% 88% and 96% 98% nucleotide sequence homology with the South Pacific and Asian isolates, respectively. The replicase encoded by DNA1 contained 186 amino acid residues, which shared 944% 958% and 976% 98.0% amino acid homology with the South Pacific and Asian isolates, respectively. The coat protein gene consists of 510 nucleotides and has 92.5% nucleotide sequence homology and 98.8% amino acid sequence homology with the Australian isolate. The protoxin gene consists of 348 nucleotides, with 90.6% nucleotide sequence homology and 87.9% amino acid sequence homology with the Australian isolate. By comparing the nucleotide sequences of DNA1, CP and MP in Chinese isolates and other national isolates, it was further proved that the BBTV isolates were obviously divided into two groups: the South Pacific and Asia. However, the amino acid sequences of the CP genes of the two isolates were very similar.
