目的建立HPLC法测定大鼠血浆中胺碘酮的含量。方法采用Agilent HC-C_(18)柱,以2%三乙胺溶液(磷酸调pH至4.0)-甲醇(19:81,V/V)为流动相,检测波长为240 nm。血浆样品经乙腈沉淀蛋白,离心后取上清液氮气吹干,按外标法定量。结果胺碘酮线性范围为0.14~2.8 mg·L~(-1)(r=0.999 0),线性关系良好,定量下限为0.098 mg·L~(-1)。日内精密度(RSD)为1.71%~2.25%,日间RSD为4.75%~6.88%,提取回收率>90%。结论本法简便、灵敏、准确,重现性好,可用于大鼠体内的药动学研究,也同样满足临床血药浓度监测和人体药动学试验。
Objective To establish an HPLC method for the determination of amiodarone in plasma of rats. Methods The mobile phase was Agilent HC-C_ (18) column with 2% triethylamine (pH adjusted to 4.0 by phosphoric acid) -methanol (19:81, V / V). The detection wavelength was 240 nm. Plasma samples were precipitated protein by acetonitrile, the supernatant was centrifuged to dry nitrogen, according to external standard method. Results The linear range of amiodarone was 0.14-2.8 mg · L -1 (r = 0.999 0). The linear range was good. The lower limit of quantitation was 0.098 mg · L -1. The intra-day precision (RSD) was 1.71% -2.25%, the intraday RSD was 4.75% -6.88%, and the extraction recovery was> 90%. Conclusion This method is simple, sensitive, accurate and reproducible. It can be used in pharmacokinetic studies in rats and also meets the clinical monitoring of plasma concentration and pharmacokinetics.
