目的:探讨含雄黄血清对星形胶质细胞(AC)兴奋性氨基酸转运体1(GLAST)、兴奋性氨基酸转运体2(GLT-1)和谷氨酰胺合成酶(GS)蛋白表达的影响。方法:以雄黄灌胃大鼠的血清为含药血清,以原代培养AC为实验对象,在总砷浓度分别为0、5、10、15、20、25μmol/L的含药血清培养基中培养48 h。采用四甲基偶氮唑盐(MTT)法检测细胞活力,Western blot法检测蛋白表达。结果:与对照组相比,AC活力、GLAST和GLT-1蛋白的表达随含雄黄血清中砷浓度的升高而逐渐降低。其中,25μmol/L含雄黄血清暴露组与对照组比较显著降低。结论:含雄黄血清暴露可对AC活力产生损伤作用,对GLAST和GLT-1蛋白表达均有明显抑制作用。
OBJECTIVE: To investigate the effect of serum containing male Huang on the expression of GLUT, GLT-1 and GS in astrocytes (AC). Methods: The serum of male rats fed with gavage was taken as the serum containing the primary cultured AC. In the medium containing 0, 5, 10, 15, 20, 25μmol / L total arsenic, Cultured for 48 h. Cell viability was detected by MTT assay and Western blot was used to detect protein expression. Results: Compared with the control group, the expression of ACTIVITY, GLAST and GLT-1 protein gradually decreased with the increase of arsenic concentration in the serum containing rye. Among them, 25μmol / L with real yellow serum exposure group was significantly lower than the control group. CONCLUSION: Serum-containing serum can damage AC activity and inhibit GLAST and GLT-1 protein expression.
