我们在进行化妆品粪大肠菌群检验时,从一份粪大肠菌群阴性,但有细菌明显生长的发酵管中分离出一株细菌。经系统生化鉴定,确认该菌为一株肺炎克雷伯氏菌。现报告如下:1 培养特性:该菌在SS、EMB平板上生长良好。菌落3—5mm,淡红色、光滑、湿润,边缘整齐;粘稠,取菌环不易挑起,可拉成丝状。在血平板上,菌落较大,灰白色,不溶血。革兰氏染色为革兰氏阴性短粗杆菌。2 生化特性:符合肠杆菌科定义:在KCN培养基及西蒙氏枸缘酸盐琼脂能生长;不液化明胶,不产H_2S,无动力;MR、苯丙氨酸脱氨酶、鸟氨酸脱羧酶、精氨酸双水解酶阴性;尿素酶、靛基质、VP赖氨酸脱羧酶阳性;分解葡萄糖产酸产气,并可迅速分解乳糖及蔗糖;该菌在液体糖、醇发酵管中,24h即可产酸,但48h后又可使培养基变碱。3 小白鼠毒力实验:取该菌肉汤培养物0.5ml注射小白鼠腹腔。4h后实验小鼠即出现躁幼不安、竖毛、卷缩震颤。8h开始出现死亡;24h内全部死亡.对照组小鼠正常。 4、小结:肺炎克雷伯氏菌在自然界水、土壤中广泛存在,亦存在于正常人的呼吸道及肠道中,是总大肠菌群的成员之一。该菌可引起大叶性肺炎、腹泻等疾病。从肺炎、腹泻等患者的各种标本中早有检出该菌的报导,但从化妆品中检出该菌尚属首例。小白鼠毒力实验证明,本次分离出的这株肺炎克?
When we tested cosmetic fecal coliforms, we isolated a strain of bacteria from a fermentation tube that was negative for fecal coliforms, but had significant bacterial growth. The system biochemical identification confirmed that the bacterium was a Klebsiella pneumoniae. The report is as follows: 1 Culture characteristics: The bacteria grew well on SS and EMB plates. Gram stain is Gram-negative Bacteroides. 2 Biochemical characteristics: in accordance with the definition of enterobacteriaceae: growth in KCN medium and Simon’s marginal acid salt agar; no liquefaction of gelatin, no production of H2S, no power; MR, phenylalanine deaminase, ornithine decarboxylation Negative for enzymes and arginine dihydrolase; positive for urease, strontium substrate, and VP-lysine decarboxylase; break down glucose to produce acid production gas, and can rapidly decompose lactose and sucrose; the bacteria is in liquid sugar and alcohol fermentation tubes, Acid production can be achieved at 24 h, but the medium can be made alkaline after 48 h. 3 Mice toxicity test: 0.5 ml of the broth culture was injected into the abdominal cavity of mice. After 4 hours, the mice developed trepidation, piloerection, and tremor. Death occurred at 8 h; all died within 24 h. Control mice were normal. 4. Summary: Klebsiella pneumoniae is widely found in water and soil in nature and is also present in the respiratory and intestinal tracts of normal people. It is one of the members of the total coliform group. The bacteria can cause diseases such as lobar pneumonia and diarrhea. A report of the bacteria was detected early in various specimens of patients with pneumonia and diarrhea, but the first case of detection of the bacteria was found in cosmetics. The toxicity test of mice showed that this strain of pneumonia was isolated this time.
