目的 :研究Graves’病 (Graves’disease ,GD)甲状腺自身抗体的异质性。方法 :以正常、GD及桥本氏甲状腺炎患者 (HT)甲状腺组织作为抗原 ,运用Westernblot检测 2 0例GD甲亢期及 2 0例GD缓解期患者的甲状腺自身抗体 ,并以正常阴性血清及标准阳性血清 [甲状球蛋白 (TGAb) +、甲状腺过氧化物酶抗体 (TPOAb) +、TSH受体抗体 (TRAb) +]作为对照。结果 :GD血清可与多种甲状腺抗原反应 ,每个患者出现的阳性条带带型均不一样。GD血清在HT及GD甲状腺组织抗原泳道出现的阳性条带数明显多于正常甲状腺组织抗原泳道出现的阳性条带数 (P <0 .0 1) ,且有的条带不出现于正常甲状腺组织抗原泳道。与标准阳性血清出现的阳性条带相比 ,GD血清出现更多的条带。结论 :GD甲状腺自身抗体存在异质性 ,GD时存在除TGAb、TPOAb、TRAb以外其它的甲状腺抗体 ,甲状腺细胞中某些蛋白的变异可能是导致GD时产生甲状腺自身抗原的原因之一。
Objective: To study the heterogeneity of thyroid autoantibodies in Graves’ disease (GD). Methods: Thyroid gland in patients with normal, GD and Hashimoto’s thyroiditis (HT) was used as antigen. Thyroid autoantibodies in 20 patients with GD hyperthyroidism and 20 patients with GD remission were detected by Western blot. Normal negative serum and standard Positive sera [TGAb +, TPOAb +, TSH receptor antibody (TRAb) +] served as controls. Results: GD serum reacted with many kinds of thyroid antigens, and the appearance of positive bands in each patient was different. GD serum in HT and GD thyroid antigen bands appeared significantly more than the normal thyroid antigen antigen band appeared in the number of positive bands (P <0.01), and some bands do not appear in normal thyroid tissue Antigen lane. The GD serum showed more bands than the positive bands that appeared in the standard positive sera. Conclusion: There is heterogeneity of thyroid autoantibodies in GD. There are other thyroid antibodies in GD except TGAb, TPOAb and TRAb. The variation of some proteins in thyroid cells may be one of the causes of thyroid autoantigen in GD.
