为了探讨牦牛AP-1基因的生理功能与结构,试验采用RT-PCR方法克隆牦牛AP-1基因,并且用多种生物信息学软件分析基因序列。结果表明:获得麦洼牦牛AP-1基因长496 bp,包含1个475 bp的开放阅读框,编码158个氨基酸。系统进化树结果显示,麦洼牦牛AP-1序列与野牦牛亲缘关系最近。AP-1编码蛋白质的相对分子质量为18.62 ku,理论等电点为5.20,不含跨膜区,不存在信号肽,二级结构以α-螺旋为主。该基因的三级结构与人AP-1蛋白质相似度高达98.05%。
In order to investigate the physiological function and structure of the yak AP-1 gene, the yak AP-1 gene was cloned by RT-PCR and analyzed by a variety of bioinformatics softwares. The results showed that AP-1 gene was 496 bp in length and contained a 475 bp open reading frame encoding 158 amino acids. The results of phylogenetic tree showed that AP-1 sequence of Maiwa yak was the closest to wild yak. The relative molecular mass of AP-1 encoded protein was 18.62 ku, the theoretical isoelectric point was 5.20, without transmembrane region and no signal peptide existed, and the secondary structure was dominated by α-helix. The tertiary structure of the gene and human AP-1 protein similarity as high as 98.05%.
