目的:探讨强骨宝方对体外高糖培养成骨细胞增殖功能的影响。方法:采用胰蛋白酶-Ⅱ型胶原酶消化法从1-2日龄SD大鼠颅盖骨中分离出成骨细胞,鉴定细胞后,用不同浓度的强骨宝方提取液及不含强骨宝方提取液的α-MEM培养液加入到成骨细胞高糖(糖终浓度为300mg/dl)培养体系,作用不同时间,应用噻唑蓝比色试验法(MTT法)(用波长490nm处OD值表示)等来检测其对成骨细胞增殖能力的影响。结果:100、50、10μg/ml的强骨方提取液对体外高糖培养成骨细胞早期的增殖功能具有明显的促进作用(P<0.05),在较高浓度时对成骨细胞增殖,无明显抑制效应。结论:强骨方提取液能促进体外高糖培养成骨细胞的增殖功能,以100、50μg/ml浓度最为合适。
Objective: To investigate the effect of QGG on proliferation of cultured osteoblasts in vitro. Methods: Osteoblasts were isolated from the calvaria of 1 - 2 day old SD rats by trypsin - Ⅱ collagenase digestion.
