研究葛根素对脑微血管内皮细胞增殖及粘附分子 ICAM- 1表达的影响 ,为脑血管疾病的病理过程提供理论依据。不同浓度的葛根素与内皮细胞孵育 2 4 h后 ,采用 MTT法检测细胞的活度 ;用 TNF- α和 IL- 1 β诱导培养的 SD大鼠脑微血管内皮细胞 ,和以葛根素处理后的内皮细胞 ,再用 TNF- α和 IL- 1 β诱导 ,采用 CEL L- EL ISA法检测粘附分子 ICAM- 1的表达。结果表明 ,TNF-α和 IL- 1 β可提高 ICAM- 1的表达 ;葛根素促进内皮细胞增殖且可抑制TNF- α和 IL- 1 β诱导的粘附分子表达。提示葛根素对防治脑血管病形成的影响因素有一定作用
To study the effect of puerarin on the proliferation of brain microvascular endothelial cells and the expression of adhesion molecule ICAM-1, and provide a theoretical basis for the pathological process of cerebrovascular diseases. Different concentrations of puerarin and endothelial cells were incubated for 24 hours. The activity of cells was detected by MTT assay; SD rat brain microvascular endothelial cells were induced with TNF-α and IL-1 β, and treated with puerarin. Endothelial cells were induced by TNF-α and IL-1β. The expression of adhesion molecule ICAM-1 was detected by CEL L-EL ISA. The results showed that TNF-α and IL-1 β can increase the expression of ICAM-1. Puerarin promotes the proliferation of endothelial cells and inhibits the expression of adhesion molecules induced by TNF-α and IL-1 β. Tips Puerarin has a role in the prevention of cerebrovascular disease formation factors
