目的探讨丙酮酸钠(SP)对红细胞储存损伤的影响。方法戊巴比妥钠腹腔注射麻醉小鼠,同时机械通气,通过心脏穿刺取血,CPDA-1作为抗凝剂和储存液,过滤去除白细胞后平分2组(n=8),SP组:添加SP浓缩液使储存红细胞中SP的终浓度为2.5 mmol/L;生理盐水(NS)组:按相同体积比加入等量的生理盐水。400 g离心15min,调整Hct为70%-75%,4℃储存14 d后,用全自动血细胞计数仪测定储存红细胞的血常规;分别用三磷酸腺苷(ATP)、丙二醛(MDA)测定试剂盒测定储存红细胞中ATP、MDA水平。结果储存14 d时,2组血细胞计数各指标无明显差异(P>0.05),且均在正常范围。NS与SP组红细胞中ATP含量(nmol/mg)为:2.46±0.19 vs 2.82±0.21(P<0.05);MDA含量(nmol/mg)为15.45±0.33 vs 14.66±0.87(P<0.05)。结论在红细胞储存过程中,SP明显提高了储存红细胞ATP水平、降低了MDA水平,具有改善红细胞储存损伤的作用。
Objective To investigate the effect of sodium pyruvate (SP) on erythrocyte storage injury. Methods The mice were anesthetized by intraperitoneal injection of pentobarbital sodium, mechanically ventilated and perfused by cardiac puncture. CPDA-1 was used as anticoagulant and storage solution. The leukocytes were separated by filtration and divided into two groups (n = 8) SP concentrate to make the final concentration of SP in the storage erythrocytes 2.5 mmol / L; NS group: the same volume of saline was added in the same volume. After being centrifuged at 400 g for 15 min, Hct was adjusted to 70% -75%, and stored at 4 ℃ for 14 days. The blood routine of erythrocytes was determined by automatic hemocytometer. The levels of ATP, MDA, The levels of ATP and MDA in stored erythrocytes were measured. Results When stored for 14 days, there was no significant difference between the two groups (P> 0.05), and they were within the normal range. The content of nmol / mg in erythrocytes of NS and SP group was 2.46 ± 0.19 vs 2.82 ± 0.21 (P <0.05). The content of MDA (nmol / mg) was 15.45 ± 0.33 vs 14.66 ± 0.87 (P <0.05). Conclusion In the process of erythrocyte storage, SP significantly increased the level of ATP in stored erythrocytes, reduced the level of MDA, and improved the storage injury of erythrocytes.
