目的:确定石荠苧总黄酮的大孔树脂吸附纯化工艺条件及其体外抑菌活性。方法:采用静态吸附和解吸实验考察不同型号大孔树脂对石荠苧总黄酮的吸附和解吸性能,确定其最佳分离纯化条件。采用二倍稀释法,检测石荠苧总黄酮对7种常见致病菌的抑菌效果。结果:HPD300大孔树脂对石荠苧总黄酮的吸附量最大,吸附率和回收率也最高。在室温下,将浓度为5.55 mg/mL(pH=3.0)的石荠苧总黄酮提取液,以2.0 mL/min速上经预处理后的HPD300大孔树脂柱,吸附后先以80 mL蒸馏水洗脱树脂柱,再用70 mL 60%乙醇(pH=8.0)以2.0 mL/min流速洗脱。此工艺条件下石荠苧总黄酮的回收率达89.12%,纯度为90.77%。纯化所得的石荠苧总黄酮对金黄色葡萄球菌、表皮葡萄球菌、痢疾杆菌具有较强的抑菌作用,其MIC分别为0.31 mg/mL、0.63 mg/mL、0.63 mg/mL。结论:HPD300大孔树脂适用于石荠苧总黄酮的分离纯化。石荠苧总黄酮对金黄色葡萄球菌、痢疾杆菌具有较强的抑菌作用。
OBJECTIVE: To determine the adsorption and purification conditions of macroporous resin and its antibacterial activity in vitro of total flavones of Mosla henryi. Methods: Adsorption and desorption properties of total flavonoids from different types of macroporous resins were investigated by static adsorption and desorption experiments, and the optimum conditions for their separation and purification were determined. The double dilution method was used to test the antibacterial effect of total flavonoids of Mosla henryi on seven common pathogenic bacteria. Results: HPD300 macroporous resin had the largest adsorption amount of total flavonoids, and the highest adsorption and recovery rate. At room temperature, a total of 5.55 mg / mL (pH = 3.0) of the total flavonoids extract of Mosla henrylum at 2.0 mL / min speed on the pretreated HPD300 macroporous resin column, after adsorption with 80 mL of distilled water The resin column was eluted and eluted with 70 mL of 60% ethanol (pH = 8.0) at a flow rate of 2.0 mL / min. Under this condition, the recovery rate of total flavonoids of Mosla henryi reached 89.12% and the purity was 90.77%. The purified total flavonoids of Mosla henryi have strong antibacterial activity against Staphylococcus aureus, Staphylococcus epidermidis and Shigella dysenteriae, and their MICs are 0.31 mg / mL, 0.63 mg / mL and 0.63 mg / mL, respectively. Conclusion: HPD300 macroporous resin is suitable for the isolation and purification of total flavonoids in Mosla.
