目的:探讨宫颈癌脆性组氨酸三联体基因(FHIT)蛋白表达与患者临床病理指标、放疗疗效及无瘤生存时间的关系。方法:用免疫组化法检测50例ⅡB~ⅢB期宫颈癌放疗前、DT10Gy,DT30Gy放疗中和放疗后FHIT蛋白的表达。结果:(1)放疗前50例患者FHIT蛋白表达阳性率36%,与病理分级,淋巴结转移有关,与年龄、临床分期、肿瘤大小无关;(2)放疗DT10Gy,DT30Gy和放疗结束后50例患者FHIT蛋白表达阳性率分别为56%,68%和84%,比放疗前明显升高(P<0.05);(3)放疗前FHIT蛋白阳性表达和阴性表达患者的放疗疗效分别为88.8%和59.4%,差异有统计学意义(P<0.05);(4)FHIT阳性表达无瘤生存时间与FHIT阴性表达无瘤生存时间无显著差异。结论:FHIT基因在宫颈癌发生发展中起重要作用,放疗可引起FHIT表达增加,FHIT蛋白的表达有助于判断宫颈癌患者放疗的敏感性。
Objective: To investigate the relationship between the expression of FHIT protein and clinicopathological parameters, radiotherapy efficacy and tumor-free survival time in cervical cancer. Methods: Immunohistochemical method was used to detect the expression of FHIT protein in 50 patients with stage ⅡB ~ ⅢB cervical cancer before radiotherapy, DT10Gy, DT30Gy radiotherapy and radiotherapy. Results: (1) The positive rate of FHIT protein in the 50 patients before radiotherapy was 36%, which was related to the pathological grade and lymph node metastasis, but not to the age, clinical stage and tumor size. (2) The radiotherapy DT10Gy, DT30Gy and 50 patients The positive rate of FHIT protein expression was 56%, 68% and 84% respectively, which was significantly higher than that before radiotherapy (P <0.05). (3) The radiotherapy effects of FHIT protein positive expression and negative expression before radiotherapy were 88.8% and 59.4 %, The difference was statistically significant (P <0.05); (4) There was no significant difference between the FHIT positive expression tumor-free survival time and FHIT negative expression tumor-free survival time. Conclusion: The FHIT gene plays an important role in the development of cervical cancer. Radiotherapy can induce the increase of FHIT expression. The expression of FHIT protein may be helpful to evaluate the sensitivity of radiotherapy in patients with cervical cancer.
