目的观察肝癌,正常肝及胎肝组织中性糖脂的不同表达及肝癌组织岩藻糖化中性糖脂的特征,并了解其对人体细胞免疫功能的影响.方法取正常肝1例,胎肝3例及肝癌4例,将标本绞碎后,经DEAEsephadexA25层析柱,碱水解和Florisil层析等方法分离和纯化中性糖脂.高效薄层色谱显色和以单波长610nm扫描分析中性糖脂,记录扫描图象和中性糖脂各组分的百分含量,并用LCAABC法进行岩藻糖基化分析.按常规方法制备NK细胞和LAK细胞,取K562细胞和肝癌Bel7402细胞分别作为NK和LAK细胞的靶细胞.用LDH释放法,设自然释放管和最大释放管.肝癌中性糖脂按4,8和12μg3个剂量梯度加样,效应细胞与靶细胞反应前先与肝癌中性糖脂反应1h.孵育18h后取上清测定光密度,计算细胞杀伤活性,用t检验进行统计学分析.结果高效薄层色谱中性糖脂显色和薄层扫描结果可见各标本中性糖脂条带显色清楚,扫描图象清晰,发现正常肝组织中性糖脂主要以多糖链(Glob)和三糖链(CTH)为主,双糖链(CDH)和单糖链(CMH)表达较低,尤其是CMH几乎缺失;胎肝CMH成低表达,而CDH表达成轻度增加;肝癌组织CDH和CMH表达明显增加,多?
Objective To observe the different expression of neutral glycolipids in hepatocellular carcinoma, normal liver and fetal liver tissues, and the characteristics of fucosylated neutral lipids in hepatocellular carcinoma tissues, and to understand their effects on human cellular immunity. Methods 1 case of normal liver, 3 cases of fetal liver and 4 cases of hepatocellular carcinoma were harvested. After the samples were minced, DEAEsephadex A25 column, alkali hydrolysis and Florisil chromatography were used to separate and purify neutral lipids. High-performance thin-layer chromatography was used for color development and the neutral glycolipids were scanned at a single wavelength of 610 nm. The percentages of the scanned images and components of neutral glycolipids were recorded. Fucosylation analysis was performed using LCA-ABC method. The NK cells and LAK cells were prepared according to conventional methods. K562 cells and hepatoma Bel7402 cells were taken as target cells for NK and LAK cells, respectively. With LDH release method, set the natural release tube and the maximum release tube. Liver cancer neutral lipids were loaded with 4, 8 and 12 μg of 3 dose gradients, and effector cells reacted with target cells before reacting with liver cancer neutral lipids for 1 h.
