目的 :探讨TNFα、Bcl- 2在大肠癌中的表达及与肿瘤大小、病理分级、临床分期和预后的关系。方法 :采用 95年前大肠癌组织存档蜡块 6 0例、腺瘤 30例、正常粘膜组织 2 0例。采用免疫组化S -P法对标本切片进行染色。结果 :TNFα在癌组织中的表达明显高于腺瘤组织和正常组织P <0 .0 1,TNFα在癌组织中的表达与Dukes分期 (A ,B期 )、五年生存期有关P <0 .0 1。Bcl- 2在腺瘤组织中表达明显高于癌组织和粘膜组织P <0 .0 1,淋巴转移组亦呈明显高表达P <0 .0 1,并且与Dukes分期 (C ,D期 )相关P <0 .0 5。TNFα与Bcl- 2在癌组织中呈负相关表达P <0 .0 1。结论 :TNFα与Bcl- 2基因参与了肿瘤的早期生物学行为 ,其检测可为大肠癌病人的生物学治疗提供依据及筛选适当的病人。
Objective: To investigate the expression of TNFα and Bcl-2 in colorectal carcinoma and its relationship with tumor size, pathological grade, clinical stage and prognosis. Methods: Sixty cases of colorectal cancer tissues, 30 cases of adenomas and 20 cases of normal mucosa tissues were collected 95 years ago. Specimens were stained with immunohistochemical S-P method. Results: The expression of TNFα in cancer tissues was significantly higher than that in adenomas and normal tissues (P <0.01). The expression of TNFα in cancer tissues was related to the Dukes stage (A and B stages) and the five year survival stage (P <0) .0 1. The expression of Bcl-2 in adenoma was significantly higher than that in carcinoma and mucosa (P <0.01). The expression of Bcl-2 was also significantly higher in P <0.01 than in Dukes stage (C and D) P <0. 0 5. TNFα and Bcl-2 in cancer tissue was negatively correlated (P <0.01). CONCLUSION: The TNFα and Bcl-2 genes are involved in the early biological behavior of tumor. The detection of TNFα and Bcl-2 gene may provide the basis for the biological treatment of patients with colorectal cancer and screen the appropriate patients.
