目的:采用半定量法检测RUNX3和EZH2在肾透明细胞癌组织中的表达,分析其相关性并探讨与肾透明细胞癌临床分期和病理分级之间的关系。方法:采用免疫组化SP法检测55例肾透明细胞癌标本和15例正常肾组织标本中RUNX3和EZH2的表达。结果:①RUNX3和EZH2在肾透明细胞癌中表达与年龄、性别差异无统计学意义(P>0.05);②EZH2在肾透明细胞癌组织标本中的表达高于正常肾组织(69.1%vs33.3%),差异有统计学意义(P<0.05);EZH2的表达随肾透明细胞癌的病理分级的升高(47.4%vs80.6%,P<0.05)和临床分期的升高(60.0%vs85.0%)而增强。③RUNX3在肾透明细胞癌组织标本中的表达低于正常肾组织(38.2%vs86.7%),差异有统计学意义(P<0.05);RUNX3的表达随肾透明细胞癌的病理分级的升高(63.2%vs25.0%,P<0.05)和临床分期的升高(51.4%vs15.0%,P<0.05)而降低。④EZH2和RUNX3在肾透明细胞癌中的表达存在负相关(r=-0.434 2,P<0.05)。结论:①RUNX3和EZH2在肾透明细胞癌组织中表达呈负相关性,并在肾透明细胞癌的发生、浸润和转移中发挥重要作用。②同时检测RUNX3和EZH2在肾透明细胞癌中的表达有助于早期诊断肾透明细胞癌、判断肿瘤的恶性程度、评估患者预后并为临床治疗提供新方法和方向。
OBJECTIVE: To detect the expression of RUNX3 and EZH2 in renal clear cell carcinoma tissues by semi-quantitative analysis, and to analyze the relationship between RUNX3 and EZH2 and the clinical stage and pathological grade of clear cell renal cell carcinoma. Methods: The expressions of RUNX3 and EZH2 in 55 cases of clear cell renal cell carcinoma and 15 cases of normal renal tissue were detected by immunohistochemical SP method. Results: ① The expression of RENX3 and EZH2 in renal clear cell carcinoma was not significantly different with age and sex (P> 0.05). ② The expression of EZH2 in renal clear cell carcinoma tissues was higher than that in normal renal tissues (69.1% vs33.3% ), The difference was statistically significant (P <0.05). The expression of EZH2 was positively correlated with the grade of renal clear cell carcinoma (47.4% vs80.6%, P <0.05) and the clinical stage (60.0% vs85. 0%). ③ The expression of RUNX3 in renal clear cell carcinoma tissues was lower than that in normal renal tissues (38.2% vs 86.7%, P <0.05). The expression of RUNX3 was positively correlated with the pathological grade of clear cell renal cell carcinoma (63.2% vs25.0%, P <0.05) and clinical stage (51.4% vs15.0%, P <0.05). ④ There was a negative correlation between the expression of EZH2 and RUNX3 in renal clear cell carcinoma (r = -0.434 2, P <0.05). Conclusion: (1) The expression of RUNX3 and EZH2 in renal clear cell carcinoma tissue is negatively correlated, and plays an important role in the occurrence, invasion and metastasis of renal clear cell carcinoma. ② Simultaneous detection of RUNX3 and EZH2 expression in renal clear cell carcinoma is helpful for the early diagnosis of clear cell renal cell carcinoma, to determine the malignancy of the tumor, to evaluate the prognosis of patients and to provide new methods and directions for clinical treatment.
