目的:研究内毒素对肾髓质水通道蛋白-2(AQP2)的影响及生长激素对AQP2表达的调控作用。方法:72只成年雄性Wistar大鼠随机分为内毒素组(予内毒素100μg/100g腹腔注射)、生长激素预处理组(予内毒素前3日每日予重组人生长激素0.1IU/100g皮下注射)和空白对照组(予生理盐水1ml腹腔注射)各24只,分别于处理后4、8、12、24h分批处死,取肾髓质石蜡切片采用免疫组化法测定AQP2表达情况。结果:与对照组相比,内毒素组AQP2表达在处理后4~8h下降20.3%、8~12h下降37.1%、12~24h下降59.8%,差异显著(P<0.05);生长激素预处理组在同时间段的AQP2表达亦明显下降(P<0.05),但较内毒素组又显著升高(P<0.05)。结论:内毒素可使肾髓质AQP2表达下调;而生长激素能够抑制内毒素对AQP2表达的影响,使肾髓质AQP2表达升高,从而对内毒素致肾损伤有一定的保护作用。
Objective: To study the effects of endotoxin on renal medulla aquaporin-2 (AQP2) and the regulatory effect of growth hormone on AQP2 expression. Methods: Seventy-two adult male Wistar rats were randomly divided into endotoxin group (100μg / 100g for endotoxin) and pretreatment with growth hormone (Control group) and blank control group (intraperitoneal injection of normal saline 1ml), 24 rats in each group were sacrificed at 4, 8, 12 and 24 hours after treatment respectively. The expression of AQP2 was detected by immunohistochemistry. Results: Compared with the control group, the AQP2 expression in endotoxin group decreased by 20.3% at 4 ~ 8 h, decreased by 37.1% at 8 ~ 12 h and decreased by 59.8% at 12 ~ 24h (P <0.05), while the growth hormone pretreatment group The expression of AQP2 was also significantly decreased in the same period (P <0.05), but significantly higher than that in the endotoxin group (P <0.05). CONCLUSION: Endotoxin can down-regulate the expression of AQP2 in renal medulla; while growth hormone can inhibit the effect of endotoxin on AQP2 expression and increase the expression of AQP2 in renal medulla, which may have a protective effect on endotoxin-induced renal injury.
