用Northern杂交、免疫细胞化学和电镜技术,以垂体ACTH瘤细胞系(AtT20)为对照,从转录、翻译及亚细胞水平检测了5种垂体外不同病理组织来源的肿瘤细胞系ACTH基因表达,包括属于APUD系统的两种人小细胞肺癌(SCLC-H69和SCLC),3种非APUD系统肿瘤:肺腺癌(AGZY83-α、低分化胃粘液癌(MGC-803)和肝癌(Bel-7402)。结果证实,(1)ACTH基因表达不仅存在于APUD系统肿瘤,而且也存在于非APUD系统肿瘤,如AGZY83-α和MGC-803。(2)不同细胞系中POMC(ACTH前体)转录的mRNA表达长度不同(800bp、1200bp、1400bp)。支持激素类物质合成在组织中普遍存在的假说,但存在着质的差别
By using Northern blot, immunocytochemistry and electron microscopy techniques, pituitary ACTH tumor cell line (AtT20) was used as a control, and the expression of ACTH genes in tumor cell lines derived from five different pituitary tissues in vitro was examined from the transcription, translation, and subcellular levels. Two human small cell lung cancers (SCLC-H69 and SCLC) of the APUD system, three non-APUD system tumors: lung adenocarcinoma (AGZY83-α, poorly differentiated gastric mucinous carcinoma (MGC-803), and liver cancer (Bel-7402) The results confirm that (1) ACTH gene expression is not only present in APUD system tumors, but also in non-APUD system tumors, such as AGZY83-α and MGC-803.
