目的探讨IFN-γ经p21信号通路调节小鼠颗粒细胞凋亡机制的初步研究。方法体外分离培养小鼠原代颗粒细胞,给予IFN-γ1000U/mL处理,在第1d和3d时,通过观察细胞形态、Real time PCR和Western Blot检测颗粒细胞p53和p21表达情况。结果镜下观察IFN-γ处理的颗粒细胞形态未见明显变化。Real time PCR和Western Blot结果示1d颗粒细胞p21表达未见明显升高,p53表达显著升高(P<0.05),而3d时颗粒细胞p21和p53表达明显升高(P<0.05)。结论 IFN-γ可能通过p53依赖的方式影响颗粒细胞p21通路的信号转导,从而导致颗粒细胞凋亡。
Objective To investigate the mechanism of IFN-γ regulated by p21 signaling pathway in mouse granulosa cell apoptosis. Methods Primary mouse granulosa cells were isolated and cultured in vitro. The cells were treated with IFN-γ1000 U / mL. The expression of p53 and p21 in granulosa cells was detected by Real Time PCR and Western Blot at 1d and 3d. Results The morphological changes of granulosa cells treated with IFN-γ were not observed under microscope. The results of Real time PCR and Western Blot showed that the expression of p21 and granulosa cells in 1d granulosa cells was not significantly increased (P <0.05), while the expression of p21 and p53 in granulosa cells was significantly increased in 3d (P <0.05). Conclusion IFN-γ may affect the signal transduction of p21 pathway in granulosa cells through p53-dependent manner, leading to the apoptosis of granulosa cells.
