应用地高辛配基标记探针和DNA-RNA原位杂交技术,检测了鼻咽部的癌组织、癌旁粘膜和慢性炎症粘膜组织中C-erbB-2,EGFRmRNA的表达。结果显示:①C-erbB-2和EGFRmRNA阳性表达率在癌组织分别为86.7%(26/30)和83.3%(25/30);癌旁粘膜分别为80.0%(16/20)和70.0%(14/20);慢性炎症粘膜均无表达。②二基因同时表达者在癌组织中有25例(83.3%),癌旁粘膜有14例(70%)。③癌旁粘膜不典型增生中二基因表达均明显高于单纯性增生;而且,轻、中、重度不典型增生的二基因阳性细胞数量逐渐增多,由上皮基底层弥散至上皮全层。提示:C-erbB-2和EGFRmRNA基因激活在鼻咽癌多阶段癌变过程是一个较早期事件,在鼻咽癌发生和发展中可能起协同作用。检测其基因表达对早期诊断鼻咽癌有一定的价值
The expression of C-erbB-2 and EGFR mRNA in nasopharyngeal carcinoma tissues, paracancerous mucosa and chronic inflammatory mucosa tissues was detected by digoxigenin-labeled probe and DNA-RNA in situ hybridization. The results showed that: (1) The positive rates of C-erbB-2 and EGFR mRNA were 86.7% (26/30) and 83.3% (25/30) respectively in the cancer tissues; 80.0% 20) and 70.0% (14/20) respectively. No expression was found in chronic inflammatory mucosa. ② Two genes simultaneously expressed in cancer tissue in 25 cases (83.3%), adjacent mucosa in 14 cases (70%). (3) The expression of two genes in adjacent mucosa atypical hyperplasia was significantly higher than that in simple hyperplasia; moreover, the number of two gene positive cells with mild, moderate and severe atypical hyperplasia increased gradually and diffused from epithelial basal layer to full layer of epithelium. It is suggested that the activation of C-erbB-2 and EGFR mRNA may be an early event in the multi-stage carcinogenesis of nasopharyngeal carcinoma and may play a synergistic role in the occurrence and development of NPC. Detection of its gene expression in the early diagnosis of nasopharyngeal carcinoma have some value
