目的:采用聚甲基丙烯酸丁酯(PBMA)整体柱毛细管电色谱法测定冬虫夏草中胞苷和腺苷的含量。方法:采用自制的PBMA电色谱整体柱(总长为34.5 cm,有效长度26.0 cm),流动相20 mmol.L-1硼砂水溶液(pH 3.5),运行电压15kV,压力进样(6×105)Pa×0.1 m in;柱温30℃,检测波长214 nm,内标溶液100μg.mL-1甲氧苄啶(TMP)溶液(乙醇-流动相1∶1为溶剂)。结果:两核苷的在12.5~125 mg.L-1线性关系良好。检测限(S/N=3)及定量限(S/N=10)分别为:胞苷2.14,7.14 mg.L-1;腺苷1.88,6.25 mg.L-1。两核苷加样回收率在97.2%~103.5%,RSD在0.9%~2.6%。结论:该方法可用于冬虫夏草中胞苷和腺苷的含量测定。
OBJECTIVE: To determine the content of cytidine and adenosine in Cordyceps sinensis using poly (butyl methacrylate) (PBMA) monolithic capillary electrochromatography. Methods: The self-made PBMA monolithic column with total length of 34.5 cm and effective length of 26.0 cm, mobile phase of 20 mmol·L-1 borax aqueous solution (pH 3.5), operating voltage of 15 kV and pressure of 6 × 105 Pa × 0.1 m in; the column temperature was 30 ℃, the detection wavelength was 214 nm, the internal standard solution was 100 μg.mL-1 trimethoprim (TMP) solution (ethanol-mobile phase 1: 1 solvent). Results: The linear relationship between two nucleosides at 12.5 ~ 125 mg.L-1 was good. The detection limit (S / N = 3) and the limit of quantification (S / N = 10) were: cytidine 2.14 and 7.14 mg.L-1; adenosine 1.88 and 6.25 mg.L-1 respectively. The recoveries of two nucleosides were between 97.2% and 103.5% with RSD between 0.9% and 2.6%. Conclusion: This method can be used for the determination of cytidine and adenosine in Cordyceps sinensis.
