目的:通过黔产莪术油对人直肠癌SW1463细胞血管内皮生长因子(VEGF)和趋化因子(chemokine,CXC)蛋白表达的影响,探讨莪术挥发油对肿瘤血管生成的作用机制。方法:将经水蒸气蒸馏提取的黔产莪术挥发油,配制成80,120,160,200 mg·L-1不同质量浓度,干预直肠癌SW1463细胞24 h后,倒置显微镜下观察不同浓度莪术油对直肠癌SW1463细胞的形态学影响;采用免疫细胞化学和蛋白免疫印迹法(Western blot)检测直肠癌SW1463细胞中VEGF,白细胞介素-8(IL-8)和CXC趋化因子受体2(CXCR2),CXC趋化因子受体3(CXCR3)蛋白的表达。结果:经莪术油处理直肠癌SW1463细胞24 h后,与空白组比较,莪术挥发油组细胞中VEGF,IL-8,CXCR2蛋白表达量明显下调(P<0.05,P<0.01),莪术油能抑制肿瘤细胞中VEGF,IL-8,CXCR2蛋白的表达;莪术挥发油组细胞中CXCR3蛋白表达量明显上调(P<0.05,P<0.01),莪术油能促进肿瘤细胞CXCR3蛋白的表达。结论:黔产莪术油可能通过下调促血管生成相关因子的表达,上调抑制性血管生成相关趋化因子的表达,从而抑制肿瘤血管生成,起到抑制肿瘤细胞的增殖作用。
OBJECTIVE: To explore the effect of Curcuma on the expression of vascular endothelial growth factor (VEGF) and chemokine (CXC) in human rectal cancer SW1463 cells and to explore the mechanism of the effect of Curcuma oil on tumor angiogenesis. Methods: The volatile oil of Curcuma phaeocaulis extracted by steam distillation was prepared into different concentrations of 80, 120, 160 and 200 mg · L-1. After intervention of rectal cancer SW1463 cells for 24 h, the morphology of rectal cancer SW1463 cells was observed with inverted microscope The expressions of VEGF, interleukin-8 (IL-8) and CXCR2 (CXCR2) and CXC chemokines in rectal cancer SW1463 cells were detected by immunocytochemistry and Western blot. Receptor 3 (CXCR3) protein expression. Results: Compared with the blank group, the expression of VEGF, IL-8 and CXCR2 in the cells of the volatile oil of Curcuma phaeocaulis decreased significantly (P <0.05, P <0.01) The expressions of VEGF, IL-8 and CXCR2 in tumor cells were significantly increased (P <0.05, P <0.01). The expression of CXCR3 protein was enhanced in the cells treated with Curcuma oil. Conclusion: Curcuma oil from Guizhou may down-regulate the expression of angiogenesis-related factors and up-regulate the expression of chemokines related to angiogenesis, thereby inhibiting tumor angiogenesis and inhibiting the proliferation of tumor cells.
