目的 分析斯氏狸殖吸虫成虫蛋白抗原及不同缓冲液制备抗原的比较。方法 用不同缓冲液制备斯氏狸殖吸虫成虫粗制抗原,并用聚丙烯酰胺凝胶电泳(SDS-PAGE)方法对其蛋白组分进行分析。结果斯氏狸殖吸虫成虫蛋白抗原共有13条蛋白带,其中有6条带染色较深,也较宽,分子量各为64KDa,56KDa,46KDa,30KDa,26KDa,22KDa。不同缓冲液制备的斯氏狸殖吸虫成虫抗原,经SDS-PAGE后其蛋白图谱完全一致。结论斯氏狸殖吸虫成虫蛋白抗原分子量主要集中在70KDa以下。不同样品缓冲液制备的抗原在应用于SDS-PAGE时不影响其蛋白分辨率。
OBJECTIVE: To analyze the protein antigens of adult Clostridium minnows and comparison of antigen preparation by different buffers. Methods The crude antigens of adult M. gifuensis were prepared with different buffers and their protein components were analyzed by polyacrylamide gel electrophoresis (SDS-PAGE). Results A total of 13 protein bands were found in the adult protein antigens of T. gondii. Among them, six bands were darker and wider, with molecular weights of 64KDa, 56KDa, 46KDa, 30KDa, 26KDa and 22KDa respectively. The adult worms of T. gondii prepared by different buffers had the same protein profile after SDS-PAGE. Conclusion The molecular weight of adult Proteus mirabilis is mainly below 70KDa. Antigens prepared in different sample buffers did not affect their protein resolution when applied to SDS-PAGE.
