目的利用Box--Behnken结合响应面法优化白鲜皮提取工艺。方法采用4因素3水平的响应面实验设计,以乙醇浓度、料液比、提取次数及提取时间为响应因子,以白鲜皮指纹图谱共有色谱峰峰面积的综合评分为指标,得出最佳工艺条件。结果采用Design Expert 8.05软件对数据进行分析,建立综合评分与影响因子的多元二次回归方程,获得的提取白鲜皮活性成分的最佳工艺条件为:乙醇浓度82.75%,料液比13.99,提取时间49.33 min,提取次数2.55。为了实际操作方便性的需要,得出实验结果为:乙醇浓度80%,料液比14,提取时间50 min,提取次数3。结论响应面分析法可为白鲜皮有效成分的提取提供科学依据,并可为制定饮片的质量标准和临床应用提供依据。
OBJECTIVE To optimize the extraction technology of fresh white skin with Box - Behnken combined with response surface methodology. Methods The response surface experimental design of 4 factors and 3 levels was used. The ethanol concentration, the ratio of solid to liquid, the number of extraction and the extraction time were used as response factors. The comprehensive score of peak area of the common chromatographic fingerprint of fresh white skin was taken as the index, Process conditions. Results Design Expert 8.05 software was used to analyze the data to establish a multiple regression equation of comprehensive score and influencing factors. The optimum conditions for the extraction of the active ingredients of white fresh skin were ethanol concentration 82.75%, solid-liquid ratio 13.99, extraction Time 49.33 min, the number of extraction 2.55. For practical convenience, the experimental results are as follows: ethanol concentration 80%, material-liquid ratio 14, extraction time 50 min, extraction times 3. Conclusion Response surface methodology can provide a scientific basis for the extraction of the effective components of P. crispus, and provide the basis for the quality standard and clinical application of the preparation of Pieces.
